利妥昔单抗是一种抗 B 细胞表面抗原 CD20 的嵌合单克隆抗体。用于治疗恶性淋巴瘤 [1] 和各种自身免疫性疾病（如类风湿性关节炎 [2] 、肉芽肿性多血管炎 [3] 、免疫性血小板减少 [4] 、重症肌无力 [5] ）。

CD20 （也称为人类 B 淋巴细胞限制性分化抗原或 Bp35 ）是正常或恶性前 B 淋巴细胞和成熟 B 淋巴细胞上的表面抗原，但在造血干细胞、前 B 淋巴细胞、浆细胞或其他组织细胞上不表达。 CD20 支持 B 细胞免疫反应，特别是针对 T 细胞非依赖性抗原 [6] ，可能作为钙离子通道。

通过与 CD20 结合，利妥昔单抗可改善 i.a. 自然杀伤 (NK) 细胞的作用， NK 细胞可诱导利妥昔标记的 B 淋巴细胞中的细胞死亡 [7] 。因此，活 B 淋巴细胞的数量明显减少。这是淋巴治疗的目的之一。在自身免疫性疾病治疗中， B 淋巴细胞的减少导致自身抗体的减少，从而导致症状的减轻。
