原位
透射电子显微术将透射电子显微镜 (TEM) 的成像能力与某些外部激源的加载相结合以观察样品的实时动态过程。 目前,有许多种系统和样品杆可以用来对样品施加不同的外场刺激来研究在偏压、机械应变/变形、加热、冷却、气体或液体环境的引入或仅仅是强电子束辐照状况下的动态过程。
过去,各种反应和系统动力学信息可记录在录像带上。在此基础上,截屏程序的出现,能够记录低分辨率的定性视频,使您能够以视频帧率,例如以30帧/秒(fps)的速度,来可视化反应情况。
现在,随着数据传输速度和数据处理能力的提升,配备了Gatan的原位相机,您可以直接对传感器输出来进行大规模数据集的记录和管理。在一次原位实验中,利用原位相机捕获的每一帧原始、定量的数据,都能被直接存储到磁盘,并以单帧图像或可回放视频的形式进行处理。此外,各种算法或脚本(例如求和、漂移校正、像素合并等)能够对采集数据进行后处理,以提取数据中的有用部分,例如下面视频所示。Gatan原位相机附带的内建管理工具使您能够轻松的对数据进行分离和提取,从而对处理后的数据进行分析,来对加载于系统的刺激导致的效应进行定量化。这一数据采集和后处理的工作流程大大提高了一次电镜机时中能够进行的实验数量。同时这一提升也使得以前无法达到的时间与空间分辨率成为可能,亚毫秒的时间分辨率,使得对于以前来说发生太快的反应能够被采集,也使得以前不为人知的动态过程得以解析。

IS相机与DENSsolutions Wildfire加热杆在1300
°C下互相配合的稳定性。以优异空间与时间分辨率捕获到紧随温度变化发生的颗粒形貌变化过程。我们无法先验的知道到底哪个(或哪些)颗粒将发生变化,但由于OneView IS相机的大视场,这些颗粒都能够被以同样的空间分辨率同时记录下来。
主视频以10倍速率进行回放,而颗粒形状变化的回放速度为2倍。内插视频进行了放大,以显示晶格条纹的解析。

该视频进行了漂移校正、背底扣除、图像过滤与裁剪。
